促精汤对小鼠生精细胞凋亡的影响

目的:探讨促精汤对模型小鼠精液常规、性激素和精子凋亡的影响。方法:40只雄性小鼠用环磷酰胺60 mg·kg-1ip 5 d造少精、弱精症模型,并随机分为模型组、促精汤低、中、高剂量组,每组10只,另设空白对照组。造模后,空白组与模型组给予生理盐水(0.5 mL/只)灌胃;促精汤低、中、高剂量(10,5,2.5 g·kg-1)灌胃,均持续30 d。采用CASA技术检测精子质量,采用酶联免疫法检测血清性激素,采用流式细胞仪检测精子凋亡率。结果:造模后模型组小鼠各级精子密度、活力和活率均明显减少(P<0.05);促精汤各剂量组明显提高小鼠精子密度、活力和活率(P<0.05),高剂量组已与空白组小鼠无显著性差异;造模后,血清黄体生成素(LH)、睾酮(T)、雌二醇(E2)显著升高,促卵泡生成激素(FSH)、催乳素(PRL)无明显变化,促精汤各剂量组能明显降低LH,T,E2水平(P<0.05),其中高剂量组已与空白组无显著差异。造模后,模型组小鼠精子凋亡显著升高(P<0.05),促精汤各组均能降低小鼠生精细胞凋亡率,高剂量组最明显,已与空白组比较无明显差异。结论:促精汤可增加各级精子密度、增强精子活力和活率,可修复睾丸病理损伤,有降低睾丸生精细胞凋亡率的作用。 Objective: To investigate the effects of promoting essence and decoction on sperm routine, sex hormones and sperm apoptosis in model mice. Methods: 40 male mice were treated with cyclophosphamide 60 mg · kg-1 ip for 5 days to make oligozoospermia and asthenospermia model and were randomly divided into model group, , Another set of blank control group. After modeling, the rats in the blank group and the model group were given normal saline (0.5 mL / body) orally. The low, medium and high dose of Jianjing Decoction (10, 5, and 2.5 g · kg -1) were given intragastrically for 30 days. The quality of sperm was detected by CASA technique. Serum sex hormones were detected by enzyme-linked immunosorbent assay. The apoptosis rate of sperm was detected by flow cytometry. Results: The sperm density, viability and viability of all the mice in the model group were significantly decreased (P <0.05) The high dose group had no significant difference with the blank group mice. After the model was established, the levels of serum LH, T and E2 were significantly increased, follicle stimulating hormone (FSH), prolactin (PRL) had no significant change, and each dose of Jianpin decoction could obviously reduce the levels of LH, T and E2 (P <0.05). There was no significant difference between the high dose group and the blank group. After modeling, the sperm apoptosis in model group was significantly increased (P <0.05), and each group of promoting decoction could reduce the rate of apoptosis of spermatogenic cells in mice, which was the most obvious in high dose group and no significant difference compared with blank group difference. Conclusion: Zhujin Decoction can increase sperm density at all levels, enhance sperm motility and viability, repair the pathological damage of testis and reduce the apoptosis rate of testicular spermatogenic cells.
