目的:建立同时测定淡豆豉中6种异黄酮类成分的分析方法,并研究炮制工艺对淡豆豉药材中异黄酮类成分的影响。方法:自行发酵淡豆豉并采用高效液相色谱法测定其异黄酮含量,使用岛津SB-C18HPLC色谱柱(4.6 mm×150 mm,5μm);以0.2%甲酸乙腈-0.2%甲酸水溶液为流动相,梯度洗脱;采用全波长紫外检测器扫描;流速1 mL·min-1,柱温35℃。结果:淡豆豉中各异黄酮色谱峰的分离度良好,在一定范围内与峰面积积分值呈良好的线性关系(r>0.999),平均加样回收率在93.16%~109.98%,RSD均<3.76%,并发现炮制过程中苷含量逐渐降低,苷元含量逐渐升高。结论:该方法操作简便、稳定、可靠,可用于淡豆豉多指标成分的含量测定,同时证明6~8 d的发酵条件最利于苷类成分的水解,较适合淡豆豉的发酵。
Objective: To establish a method for the simultaneous determination of 6 isoflavones in Beauveria bream and to study the effect of processing technology on isoflavones in Beauveria bream. Methods: The isoflavone content was determined by high performance liquid chromatography (HPLC). The reaction was performed on a Shimadzu SB-C18 HPLC column (4.6 mm × 150 mm, 5 μm) using 0.2% formic acid in acetonitrile and 0.2% formic acid as mobile phase , Gradient elution; using full-wavelength UV detector scan; flow rate of 1 mL · min-1, column temperature 35 ℃. Results: The separation of each isoflavone peak was very good in a certain range, with a good linear relationship with the peak area integral (r> 0.999). The average recovery was 93.16% -109.98%, RSD was < 3.76%, and found that during the processing of glycosides gradually reduce the content of aglycone gradually increased. Conclusion: The method is simple, stable and reliable. It can be used to determine the content of multi-index components of Beauveria bream. It is also proved that the fermentation conditions of 6-8 days are the most suitable for the hydrolysis of glycosides, which is more suitable for the fermentation of Beauveria bream.
