本研究利用ISSR分子标记技术对22个郁金香品种进行遗传多样性分析,以期了解郁金香不同品种之间的遗传关系,提高育种的目标性。利用11条引物共扩增出129条谱带,其中多态性谱带为124条,多态性率为96.1%,22个郁金香品种的检测等位基因数(na*)平均为1.983 9,有效等位基因数(ne*)平均为1.383 3,Nei’s基因多样性指数(h*)平均为0.249 0,Shannon’s信息指数(I*)平均为0.399 0,表明郁金香多态性较高,具有丰富的遗传多样性。利用NTSYS软件进行UPGMA聚类分析,22个品种在遗传相似系数0.62为阈值处可分为两大类,根据聚类结果,理论上推测不同花色的品种间杂交获得新品种的潜力较大。此结论为进一步研究和利用郁金香提供分子水平的依据。
In this study, genetic diversity of 22 tulip cultivars was analyzed using ISSR molecular markers in order to understand the genetic relationship among different tulip cultivars and to improve the breeding target. A total of 129 bands were amplified with 11 primers, of which 124 bands were polymorphic with a polymorphism rate of 96.1%. The mean number of alleles detected (na *) of 22 tulip cultivars was 1.983 9, The average number of effective alleles (ne *) was 1.383 3, the average Nei’s gene diversity index (h *) was 0.249 0, and Shannon’s information index (I *) was 0.399 0, indicating that the tulip polymorphism was high and abundant Genetic diversity. UPGMA cluster analysis using NTSYS software, 22 varieties in the genetic similarity coefficient of 0.62 for the threshold can be divided into two categories, according to the clustering results, theoretically speculated that different varieties of hybrids to obtain new varieties of great potential. This conclusion provides the basis for further research and utilization of tulip to provide molecular level.
