目的:研究基质金属蛋白酶-2(matrix metalloproteinase 2,MMP-2)、基质金属蛋白酶-14(matrix metalloproteinase 14,MMP-14)和RECK基因在涎腺多形性腺瘤(pleomorphic adenoma,PA)初发患者、复发患者及进一步恶变患者组中的表达及其意义。方法:研究对象为新疆医科大学第一附属医院颌面肿瘤外科2004年至2014涎腺PA手术标本50例,其中初发36例,恶变7例,复发7例和10例正常腮腺组织(10例均来自腮腺)。通过免疫组织化学方法检测RECK、MMP-2和MMP-14在各组标本中的表达,并采用半定量方法进行计量。对RECK与MMP-2及MMP-14在各组中的表达采用SPSS 21.0软件包对数据进行统计学分析。结果:MMP-2在正常组与复发组差异有统计学意义(P<0.05),正常组与恶变组差异有统计学意义(P<0.05),初发组与复发组差异有统计学意义(P<0.05),初发组与恶变组差异有统计学意义(P<0.05)。RECK与MMP-2和MMP-14在各个组的表达都呈负相关(r=-0.887,P<0.05;r=-0.798,P<0.05)。结论:临床上,在涎腺PA恶变,复发组织同时对RECK与MMP-2的进行检测分析其有重要意义,这对于判别肿瘤侵蚀性和预后指标是极有可能。
OBJECTIVE: To study the effect of matrix metalloproteinase 2 (MMP-2), matrix metalloproteinase 14 (MMP-14) and RECK gene on the development of pleomorphic adenoma (PA) Patients, relapse patients and further malignant group of patients and its significance. Methods: The study was performed on 50 patients with salivary gland salivary gland from 2004 to 2014 in the First Affiliated Hospital of Xinjiang Medical University. There were 36 cases of initial salivary gland, 7 cases of malignant transformation, 7 cases of recurrence and 10 cases of normal parotid gland (10 cases All from the parotid gland). The expression of RECK, MMP-2 and MMP-14 in each group of specimens was detected by immunohistochemical method and was measured by semi-quantitative method. The data of RECK, MMP-2 and MMP-14 in each group were analyzed by SPSS 21.0 software package. Results: The difference of MMP-2 in normal group and recurrent group was statistically significant (P <0.05), the difference between normal group and malignant group was statistically significant (P <0.05), the difference between initial group and recurrent group was statistically significant P <0.05), the difference between the initial group and the malignant group was statistically significant (P <0.05). RECK was negatively correlated with the expression of MMP-2 and MMP-14 in all groups (r = -0.887, P <0.05; r = -0.798, P <0.05). Conclusion: In clinic, it is very important to detect and analyze RECK and MMP-2 in salivary gland PA malignant transformation and recurrence tissue, which is very important for discriminating tumor aggressiveness and prognosis.
