目的探讨采用高效液相色谱法测定白附片中新乌头碱、次乌头碱和乌头碱含量的应用价值。方法选用反相离子对HPLC法,采用Agilent Zorbax Eclipse XDB-C18柱(5μm,4.6×250mm);流动相:乙腈-5mmol-1Na H2PO4溶液,调整磷酸至p H(52:50),内含有6mmol-1的十二烷基磺酸钠(SDS);流速为1.0m L·min-1;检测波长为234nm。结果白附片中新乌头碱、次乌头碱和乌头碱可完全分离,且能准确测定。结论此方法具有较高准确度,在白附片中生物碱含量测定中具有良好的应用价值。
Objective To investigate the application of high performance liquid chromatography (HPLC) in the determination of mesaconitine, hypaconitine and aconitine in Baifu Tablet. Methods The mobile phase was acetonitrile-5mmol-1Na H2PO4 solution with reversed-phase ion-pair HPLC using Agilent Zorbax Eclipse XDB-C18 column (5μm, 4.6 × 250mm) -1 sodium dodecyl sulfate (SDS); the flow rate was 1.0 m L · min-1; the detection wavelength was 234 nm. Results Mesena in the new aconitine hypaconitine and aconitine can be completely separated, and can be accurately measured. Conclusion This method has high accuracy and has good application value in the determination of alkaloids in the white tablets.
