目的:通过右归丸含药血清对大鼠成骨细胞GPR48、ATF4、BSP的实验研究,探究右归丸对治疗骨质疏松症的疗效机制。从而为“肾藏精”这一理论提供实验依据。方法:将SPF级大鼠随机分为正常组、右归丸组、诱导液组、骨疏康组、福善美组,对大鼠进行灌胃5 d。制备含药血清,并观察右归丸含药血清对成骨细胞的影响。结果:与正常组和阳性药物组及诱导液组相比,右归丸组的蛋白表达水平明显上升。结论:(1)正常大鼠骨组织中存在GPR48、ATF4、BSP蛋白表达;(2)右归丸组中的GPR48、ATF4、BSP与正常组相比含量均大幅提升,说明右归丸含药血清能够调控GPR48、ATF4、BSP的蛋白浓度,并可促进骨髓间充质干细胞向骨细胞分化,从而对骨质疏松症起到防治作用。
OBJECTIVE: To investigate the therapeutic effect of Yougui Wan on osteoporosis GPR48, ATF4 and BSP in rats by using the serum containing Yougui Wan. This provides an experimental basis for the theory of “kidney essence”. Methods: SPF rats were randomly divided into normal group, Yougui Wan group, induced liquid group, Gushukang group and Fosamax group. The rats were gavaged for 5 days. Preparation of drug-containing serum, and observe Yougui pill containing serum on osteoblasts. Results: Compared with the normal group and the positive drug group and the inducing fluid group, the protein expression of Yougui pill group increased significantly. CONCLUSION: (1) GPR48, ATF4 and BSP protein expression exist in normal rat bone tissue. (2) The contents of GPR48, ATF4 and BSP in Youguiwan group increased significantly compared with the normal group, Serum can regulate the protein concentration of GPR48, ATF4, BSP, and can promote bone marrow mesenchymal stem cells to differentiate into osteoblasts, thereby preventing and treating osteoporosis.
