目的:研究不同剂量60Coγ照射大鼠腮腺组织后SSB1蛋白及基因的表达变化情况。方法:选取SD大鼠60只,随机分成6组,每组10只。对照组(未照射组)和实验组(3 Gy、6 Gy、9 Gy、12 Gy、15 Gy组),实验组于放射后6 h处死大鼠。处死后收集大鼠腮腺组织,免疫组化法检测不同剂量放射线照射后SSB1蛋白在大鼠腮腺组织中的表达情况及实时荧光定量PCR法检测不同剂量放射线照射后SSB1基因在大鼠腮腺组织中的表达情况。结果:放射后大鼠的腮腺组织中的SSB1的表达量随着放射剂量不同而变化,并且在12 Gy达到最高,实验组和对照组比较差异均有统计学意义(P<0.05)。结论:大鼠腮腺组织中SSB1蛋白及基因的表达无剂量依赖性,其表达量随放射剂量的增加先增加后降低。
Objective: To study the changes of SSB1 protein and gene expression in the parotid glands of rats exposed to different doses of 60Coγ. Methods: Sixty Sprague-Dawley rats were randomly divided into 6 groups with 10 rats in each group. Rats in the control group (non-irradiated group) and experimental group (3 Gy, 6 Gy, 9 Gy, 12 Gy and 15 Gy) were sacrificed at 6 hours after irradiation. After sacrifice, the parotid gland tissues of rats were collected. The expression of SSB1 protein in the parotid gland tissue of rats was detected by immunohistochemistry and the expression of SSB1 protein in the parotid gland tissue of rats were detected by real-time fluorescence quantitative PCR after different doses of radiation Express the situation. Results: The expression of SSB1 in the parotid gland of rats after radiotherapy varied with the dose of radiation and reached the highest at 12 Gy. The difference between the experimental group and the control group was statistically significant (P <0.05). Conclusion: The expression of SSB1 protein and gene in the parotid gland of rats has no dose-dependent manner. The expression level of SSB1 first increased and then decreased with the increase of radioactive dose.
