目的:探究益脑方改善东莨菪碱所致小鼠学习记忆障碍的作用及其机制。方法:昆明种小鼠灌胃给予益脑方(0.258、0.517、0.775g/kg)水溶液,利用东莨菪碱制备记忆障碍模型,Morris水迷宫和跳台测试小鼠学习记忆能力。ELISA法或酶分析法测定脑内Ach、AchE、DA、5-HT、NA、SOD、MDA、脂褐素(Lipo)和血浆皮质酮(CORT)的含量,Western blot法测定小鼠脑内NMDA受体1型亚基(NR1)和突触蛋白(SYN)的含量,微阵列系统测定小鼠海马神经元突触可塑性。结果:0.517和0.775g/kg益脑方能够改善小鼠水迷宫行为学实验中学习记忆能力,0.258g/kg还可改善小鼠跳台实验中的行为学表现;益脑方增加脑Ach、5-HT、NA、DA和SOD的含量;还可以降低血浆CORT及脑Ach E、MDA和Lipo含量,提高海马脑片EPSP幅度。结论:益脑方具有改善东莨菪碱所致学习记忆障碍的作用,其机制可能与其调整或调节与记忆相关的神经递质、抗氧化物质含量以及突触可塑性等有关。
Objective: To investigate the effect of YINNAN on learning and memory impairment induced by scopolamine and its mechanism. Methods: Kunming mice were orally given yinnaofang (0.258,0.517,0.775g / kg) aqueous solution, scopolamine was used to prepare memory impairment model, Morris water maze and jumping test mice learning and memory ability. The levels of Ach, AchE, DA, 5-HT, NA, SOD, MDA, Lipo and CORT in brain were measured by enzyme-linked immunosorbent assay (ELISA) Receptor type 1 subunit (NR1) and synaptophysin (SYN) were measured by microarray system. The synaptic plasticity of hippocampal neurons in mice was determined by microarray system. Results: 0.517 and 0.775g / kg of YINFANG can improve learning and memory abilities in water maze behavior test, and 0.258g / kg can also improve behavioral performance of mice in jumping table test. YINFANG increases brain Ach, 5 -HT, NA, DA and SOD; also can reduce plasma CORT and brain AchE, MDA and Lipo content, improve EPSP amplitude of the hippocampal slices. Conclusion: YIN NONGFANG can improve learning and memory impairment induced by scopolamine, and its mechanism may be related to its regulation or regulation of memory-related neurotransmitters, antioxidants and synaptic plasticity.
