目的 :探讨三氧化二砷 (AS2 O3)对肺癌细胞株NCI -H6 9的增殖抑制和诱导凋亡作用。方法 :MTT法观察不同浓度AS2 O3 作用不同时间后的NCI -H6 9生长的影响 ;增殖细胞核抗原 (PCNA)单抗LSAB免疫组化染色 -流式细胞仪检测PCNA阳性细胞百分比 ;FTIC -TUNEL法 -流式细胞仪检测细胞凋亡指数。结果 :随AS2 O3 浓度增加和作用时间延长 ,NCI -H6 9肺癌细胞的细胞活度渐下降。随AS2 O3 浓度增加 ,PCNA阳性细胞百分比逐渐下降 ,而凋亡指数逐渐升高。结论 :三氧化二砷可明显抑制肺癌细胞株NCI -H6 9的生长 ,并能诱导细胞凋亡 ,有治疗肺癌的潜在价值
Objective: To investigate the effect of arsenic trioxide (AS2O3) on proliferation inhibition and apoptosis of lung cancer cell line NCI-H6 9. METHODS: MTT assay was used to observe the effects of different concentrations of AS2O3 on the growth of NCI-H6 9 cells; Proliferating cell nuclear antigen (PCNA) monoclonal antibody LSAB immunohistochemical staining-Flow cytometry to detect the percentage of PCNA-positive cells; FTIC-TUNEL method - Apoptosis index was measured by flow cytometry. Results: The cell activity of NCI-H6 9 lung cancer cells gradually decreased with the increase of AS2 O3 concentration and the prolonged action time. As the concentration of AS2 O3 increased, the percentage of PCNA-positive cells gradually decreased while the apoptotic index gradually increased. Conclusion : Arsenic trioxide can significantly inhibit the growth of lung cancer cell line NCI-H6 9 and induce apoptosis. It has potential value in the treatment of lung cancer.
