目的:建立附桂骨痛颗粒浸膏中芍药苷的HPLC含量测定方法,为该药品中所含芍药药材提供质量控制标准。方法:以甲醇为溶剂,超声40分钟提取,滤过,滤液蒸干,50%甲醇溶解,过柱,洗脱,采用C18柱(150 mmx4.6 mm,5μm)为分析柱;流动相为甲醇-水(25∶75);检测波长为230 nm;柱温30℃。结果:芍药苷的线性范围为0.04051μg~0.6482μg(r=0.9999),线性关系良好;平均回收率为99%(RSD为1%)。结论:该方法快速简便、准确可靠,重现性好,结果稳定,可用于附桂骨痛颗粒浸膏中芍药药材的质量控制。
OBJECTIVE: To establish a HPLC method for determination of paeoniflorin in Fugu Gutong Granule Extract, and to provide quality control standards for Paeonia lactiflora in this medicine. Methods: The sample was extracted with 40% ultrasonic for 40 minutes and filtered. The filtrate was evaporated to dryness and dissolved in 50% methanol. The column was eluted with C18 column (150 mm × 4.6 mm, 5 μm). The mobile phase consisted of methanol - water (25:75); detection wavelength was 230 nm; column temperature was 30 ° C. Results: The linear range of paeoniflorin was 0.04051μg ~ 0.6482μg (r = 0.9999), and the linear relationship was good. The average recovery was 99% (RSD 1%). Conclusion: The method is rapid, simple, accurate and reliable with good reproducibility and stable results. It can be used for the quality control of Paeonia lactiflora.
