目的:研究牛磺酸对大鼠贮脂细胞、肝细胞增殖及前胶原基因表达的影响,为应用牛磺酸防治肝纤维化提供实验依据。方法:分离、培养大鼠贮脂细胞和肝细胞;采用~3H-胸腺嘧啶和~3H-脯氨酸掺入测定细胞增殖及胶原台成;用原位杂交检测Ⅰ、Ⅲ型前胶原 mRNA 含量。结果:在5~80mmol/L 浓度范围内,牛磺酸能显著抑制贮脂细胞、肝细胞的增殖及胶原合成,并呈时间、剂量依赖性关系;20mmol/L 牛磺酸可显著抑制Ⅰ、Ⅲ型前胶原 mRNA 的表达。结论:牛磺酸在体外具有抗肝纤维化作用,它对于防治肝纤维化可能有一定的临床意义和应用前景。
Objective: To study the effect of taurine on the storage of fat-storing cells, hepatocyte proliferation and procollagen gene expression in rats, and to provide experimental evidence for the prevention and treatment of hepatic fibrosis with taurine. Methods: The lipid-storing cells and hepatocytes of rats were isolated and cultured. The proliferation and collagen synthesis of cells were detected by ~ 3H-thymidine and 3H-proline incorporation. The mRNA of type Ⅰ and Ⅲ procollagen was detected by in situ hybridization . Results: Taurine significantly inhibited the proliferation and collagen synthesis of lipid-storing cells and hepatocytes in a dose-dependent manner in a concentration range of 5 ~ 80mmol / L, Type Ⅲ procollagen mRNA expression. Conclusion: Taurine has an anti-hepatic fibrosis effect in vitro. It may have some clinical significance and application prospects for the prevention and treatment of liver fibrosis.
