目的探讨Twist和E-cadherin蛋白在大肠癌发生发展中的临床意义。方法采用免疫组化PV9000法检测78例大肠癌和42例大肠腺瘤组织中Twist和E-cadherin蛋白的表达。采用Westernblot法检测Twist蛋白在30例新鲜大肠腺癌组织和30例新鲜大肠腺瘤组织中的表达。结果Twist蛋白在大肠癌组织中的阳性率(56.4%)明显高于腺瘤组织(26.2%)和癌旁正常黏膜组织(10%);E-cadherin在大肠癌组织中的阳性率(48.7%)明显低于腺瘤组织(76.2%)和癌旁正常黏膜组织(100%)。Twist和E-cadherin在大肠癌中的异常表达均与肿瘤分化程度和淋巴结转移有关(P均<0.05),而与肿瘤大小无关(P>0.05)。E-cadherin的阳性表达与肿瘤的TNM分期有关(P<0.05),但Twist的阳性表达与TNM分期无关(P>0.05)。结论Twist和E-cadherin蛋白在大肠癌的发生及癌组织的侵袭和转移过程中可能起重要作用。
Objective To investigate the clinical significance of Twist and E-cadherin in the development of colorectal cancer. Methods The expression of Twist and E-cadherin protein in 78 cases of colorectal carcinoma and 42 cases of colorectal adenoma was detected by immunohistochemistry PV9000. Western blot was used to detect the expression of Twist protein in 30 cases of fresh colorectal adenocarcinoma and 30 cases of fresh colorectal adenoma. Results The positive rate of Twist protein in colorectal carcinoma (56.4%) was significantly higher than that in adenoma (26.2%) and adjacent normal mucosa (10%). The positive rate of E-cadherin in colorectal carcinoma was 48.7% ) Was significantly lower than adenoma (76.2%) and adjacent normal mucosa (100%). The abnormal expression of Twist and E-cadherin in colorectal cancer were correlated with tumor differentiation and lymph node metastasis (all P <0.05), but not with tumor size (P> 0.05). The positive expression of E-cadherin was correlated with the TNM stage (P <0.05), but the positive expression of Twist was not related to the TNM stage (P> 0.05). Conclusion Twist and E-cadherin proteins may play an important role in the carcinogenesis and metastasis of colorectal cancer.
