目的观察并探讨5种不同灭菌方法、剂量及p H值糖(醇)培养基对不同培养液、不同浓度鼠疫菌的发酵结果,筛选最适鼠疫菌发酵的糖(醇)培养基。方法选取对鼠疫菌具有典型鉴定特征的5种糖(醇),观察制作过程中不同剂量、不同pH值和不同灭菌方法对鼠疫菌发酵结果的影响。结果不同灭菌方法、不同培养液的5种糖(醇)培养基对不同浓度的鼠疫菌发酵结果无影响(141菌株:阿胶糖(+)、鼠李糖(-)、密二糖(-)、麦芽糖(+)、甘油(+);EV 76:甘油(-),其它发酵结果同141菌株,均为正常发酵);不同剂量及pH值的糖(醇)培养基对鼠疫菌发酵结果有一定的影响,特别是剂量为0.5%、pH值为7.2~7.4的阿胶糖和甘油可引起迟缓发酵。结论制备鼠疫菌糖(醇)培养基的最适剂量为1%,pH值为7.6,是鼠疫菌发酵的理想选择值。
Objective To observe and discuss the fermentation results of five kinds of different sterilization methods, doses and pH value sugar (alcohol) medium on different culture medium and different concentrations of Yersinia pestis, and select the optimum culture medium for the fermentation of Yersinia pestis. Methods Five kinds of sugars (alcohol) with typical identification characteristics of Yersinia pestis were selected. The effects of different dosages, different pH value and different sterilization methods on the fermentation results of Y. pestis were observed. Results The results showed that there was no effect on the fermentation results of Yersinia pestis at different concentrations with different sterilization methods and different culture media (141 strains: gelatin (+), rhamnose (-), ), Maltose (+), glycerol (+); EV 76: glycerin (-), and other fermentative strains were the same as those of the strain 141). The results of fermentation of Yersinia pestis with different dosages and pH Have a certain impact, especially the dose of 0.5%, pH value of 7.2 ~ 7.4 gelatin and glycerol can cause slow fermentation. Conclusion The optimum dosage for the production of Yersinia pestis (alcohol) medium is 1% and the pH value is 7.6, which is the ideal choice for Yersinia pestis fermentation.
