目的:观察抗郁散对慢性不可预见性轻度应激抑郁症模型(CUMS)大鼠的行为学变化及其大脑海马5-HT1AR基因表达水平的影响。方法:将46只成年雄性SD大鼠,随机分为正常空白组、模型对照组、丁螺环酮组和抗郁散组4组,除正常组外其余大鼠利用CUMS方法复制抑郁模型。运用RT-PCR技术扩增,凝胶图像分析系统中对各目的基因条带的浓度进行测定。结果:抗郁散组与模型组比较,大鼠的体质量明显增加(P<0.05),水平和垂直行为活动明显增多(P<0.05),对糖水消耗明显增强(P<0.05),海马5-HT1AR基因表达显著提高(P<0.01)。结论:抗郁散具有改善CUMS大鼠行为学变化的特征,明显提高CUMS大鼠海马5-HT1AR的mRNA表达水平,提高5-HT的活性从而达到治疗抑郁症的目的。
Objective: To observe the effect of Kangyu Powder on the behavior changes and the expression of 5-HT1AR gene in the hippocampus of chronic unpredictable mild stress depression model (CUMS) rats. Methods: Forty-six adult male Sprague-Dawley rats were randomly divided into 4 groups: normal control group, buspirone group and Kangyuashan group. The rest rats were deprived of depression model by CUMS method. Using RT-PCR amplification, gel image analysis system for the purpose of the determination of the concentration of gene bands. Results: Compared with the model group, the body mass of rats in Kangyu powder group was significantly increased (P <0.05), the activities of horizontal and vertical behavior were significantly increased (P <0.05), the consumption of sugar water was significantly increased (P <0.05) -HT1AR gene expression was significantly increased (P <0.01). CONCLUSION: Kangyu powder can improve the behavioral changes of CUMS rats, increase the mRNA expression level of 5-HT1AR in the hippocampus and increase the activity of 5-HT in CUMS rats so as to achieve the goal of treating depression.
