从分泌抗乙型脑炎病毒中和抗体的杂交瘤细胞株51-8的基因文库中筛选出的功能性重链和轻链可变区基因,分别与人y~3链恒定区基因和k链恒定区基因连接,构建成人-鼠嵌合的重链和轻链基因,两种嵌合基因共转染骨髓瘤细胞Sp/0,转染率为2.8×10~(-7)。转染细胞在体外传代30代以上,其上清液能特异地与羊抗人IgG(Fc特异的)抗体反应,并能稳定地表达出抗乙型脑炎病毒的人-鼠嵌合抗体。
