目的观察信号转导和转录活化因子3(STAT3)和细胞周期素D1(Cyclin D1)在大鼠肝癌组织中的表达变化,并探讨其意义。方法将72只大鼠随机分为模型组和对照组各36只,模型组自由饮用0.1 mg/mL的二乙基亚硝胺(DEN)溶液,对照组饮用等量灭菌蒸馏水,连续饮用20周。处死大鼠取癌组织,用RT-PCR法检测STAT3、Cyclin D1 mRNA,Western blot法和免疫组化法检测STAT3、Cyclin D1蛋白。结果对照组STAT3、Cyclin D1mRNA表达水平分别为0.32±0.12、0.18±0.05,模型组分别为0.72±0.25、0.57±0.15,P均<0.05。Western blot法检测模型组STAT3、Cyclin D1蛋白表达量分别为0.58±0.25、0.65±0.14,对照组分别为0.32±0.19、0.41±0.15,P均<0.05。免疫组化法检测模型组STAT3、Cyclin D1蛋白IOD分别为2 373.87±258.79、668.44±63.10,对照组分别为487.81±55.31和138.86±31.50,P均<0.05。结论 STAT3、Cyclin D1在肝癌大鼠癌组织中表达上调,二者可促进肝癌的发生和发展。
Objective To observe the expression changes of signal transducer and activator of transcription 3 (STAT3) and cyclin D1 in rat hepatocellular carcinoma (HCC) and to explore its significance. Methods Seventy-two rats were randomly divided into model group and control group, with 36 rats in each group. The rats in the model group were given 0.1 mg / mL diethylnitrosamine (DEN) solution freely. The control group received equal amount of sterile distilled water and 20 week. The rats were sacrificed and the cancerous tissues were taken out. STAT3 and Cyclin D1 mRNA were detected by RT-PCR. Western blot and immunohistochemistry were used to detect STAT3 and Cyclin D1 protein. Results The STAT3 and Cyclin D1 mRNA expression levels in the control group were 0.32 ± 0.12 and 0.18 ± 0.05, respectively. The model group were 0.72 ± 0.25 and 0.57 ± 0.15 respectively, all P <0.05. The protein expression levels of STAT3 and Cyclin D1 in the model group were 0.58 ± 0.25 and 0.65 ± 0.14 respectively by Western blot and 0.32 ± 0.19 and 0.41 ± 0.15 respectively in the control group, P <0.05. The IOD of STAT3 and Cyclin D1 in the model group were respectively 2 373.87 ± 258.79 and 688.44 ± 63.10 by immunohistochemistry, and 487.81 ± 55.31 and 138.86 ± 31.50 respectively in the control group, P <0.05. Conclusion STAT3 and Cyclin D1 are upregulated in HCC tissues and both of them can promote the occurrence and development of HCC.
