目的观察中药脑疏宁对创伤性脑损伤(TBI)大鼠血脑屏障(BBB)通透性及脑组织基质金属蛋白酶9(MMP9)蛋白表达的影响,探讨中药保护血脑屏障的作用机理。方法按Feeney自由落体撞击法造成TBI模型,采用免疫组织化学技术观察大鼠脑组织MMP9表达。结果模型组创伤后24hBBB破坏,脑组织含水量增加,病灶区MMP9蛋白表达增高,脑疏宁组BBB通透性、脑组织含水量及MMP9表达较模型组降低(P<0.01)。结论颅脑损伤后MMP9表达增高,促使BBB破坏,导致血管源性脑水肿。中药脑疏宁可以保护BBB,降低脑组织含水量,其作用可能与抑制MMP9在损伤脑组织中的表达有关。
Objective To observe the effects of Chinese herbal medicine Naoshuning on the permeability of blood-brain barrier (BBB) and the expression of matrix metalloproteinase 9 (MMP9) in brain tissue of traumatic brain injury (TBI) rats, and to explore the protective mechanism of Chinese medicine on blood-brain barrier. Methods TIE model was induced by Feeney free-fall impaction method. The expression of MMP9 in rat brain tissue was observed by immunohistochemistry. Results BBB destruction, brain water content increased, and MMP9 protein expression increased in the model group at 24 hours after trauma. Brain permeability, brain water content, and MMP9 expression in Nashunning group were lower than those in the model group (P<0.01). Conclusion The expression of MMP9 increases after craniocerebral injury, which leads to the destruction of BBB and leads to vasogenic brain edema. The Chinese medicine brain shu shu Ning can protect BBB and reduce the water content of brain tissue, and its effect may be related to the inhibition of the expression of MMP9 in damaged brain tissue.
