目的研究温胆汤对MK-801诱发精神分裂症模型鼠海马组织PKC含量及其超微结构的影响。方法将50只清洁级SD大鼠随机分为5组:正常组(A)、模型组(B)、温胆汤高剂量组(C)、温胆汤中剂量组(D)、温胆汤低剂量组(E),每组10只。在造模前,C,D,E组连续ig温胆汤,每次给药20ml·kg-1(相当于C、D、E组剂量为生药40,20,10g·kg-1);A、B组用等量生理盐水ig,1次/d,共21d。在最后一次给药2 h后,B,C,D,E组一次性左侧腹腔ip MK-8010.6 mg·kg-1,建立精神分裂症模型;一般状况观察3 d后,处死大鼠,取海马组织检测。采用双抗体夹心-酶联免疫吸附(ABC-ELISA)法检测海马组织中PKC的含量,电镜观察海马组织的超微结构。结果与模型组比较,温胆汤各组均不同程度改善了模型鼠的一般状况,并且显著降低PKC的含量(P<0.05或P<0.01)和减轻神经元细胞的凋亡。结论温胆汤可明显减轻精神分裂症模型鼠神经元细胞的凋亡,降低PKC浓度,从而对缝隙连接通讯(GJIC)的功能起到调节作用。
Objective To study the effect of Wendan Decoction on PKC content and ultrastructure in hippocampus of MK-801-induced schizophrenia model rats. Methods Fifty clean-grade SD rats were randomly divided into five groups: normal group (A), model group (B), high-dose group of Wendan decoction (C), middle-dose group of Wendan decoction (D) Low-dose group (E), each group of 10. Results Compared with the model group, each group of Wendan Decoction improved the general condition of the model rats to varying degrees, and significantly reduced the content of PKC (P <0.05 or P <0.01) and alleviated the apoptosis of neurons. Conclusion Wen Dan Tang can significantly reduce the apoptosis of neuronal cells in schizophrenic model mice and decrease the concentration of PKC, which plays a regulatory role in the function of gap junctional communication (GJIC).
