目的 :了解食管癌细胞分化过程中DNA聚合酶 β(polβ)基因的表达。方法 :采用全反式维甲酸 (ATRA)诱导人食管癌Eca1 0 9细胞分化 ,应用甲基绿 派洛宁染色、原位杂交、免疫组化和Westernblot方法分别检测细胞分化和polβ基因的表达。 结果 :ATRA作用 5d后可以诱导Eca1 0 9细胞分化。wt polβ和mt polβ(截短的 )基因的表达随着Eca1 0 9细胞的分化而降低。结论 :ATRA可下调Eca1 0 9细胞wt polβ和mt polβ的表达 ,促使其分化
Objective: To investigate the expression of DNA polymerase β (polβ) gene during esophageal cancer cell differentiation. Methods: All-trans retinoic acid (ATRA) was used to induce the differentiation of human esophageal cancer Eca1 0 9 cells. Methyl green peony staining, in situ hybridization, immunohistochemistry and Western blot were used to detect the expression of polβ gene. Results: After treated with ATRA for 5 days, Eca109 cells could be induced to differentiate. The expression of wt polβ and mt polβ (truncated) genes decreased with the differentiation of Eca109 cells. Conclusion: ATRA can down-regulate the expression of wt polβ and mt polβ in Eca1 0 9 cells,
