目的通过检测磷酸化信号转导和转录激活因子3(p-STAT3)(Y705)和血管内皮生长因子(VEGF)在正常子宫内膜与子宫腺肌症在位内膜中的定位与表达,探讨p-STAT3(Y705)和VEGF在子宫腺肌症中发生发展中的作用。方法免疫组织化学SP法检测14例正常子宫内膜和14例子宫腺肌症在位内膜中p-STAT3(Y705)和VEGF的定位和表达情况;Real-time PCR检测正常子宫内膜与子宫腺肌症在位内膜组织中STAT3 mRNA表达情况;Western blotting检测正常子宫内膜与子宫腺肌症在位内膜组织中p-STAT3(Y705)、STAT3和VEGF蛋白的表达水平。结果 p-STAT3(Y705)主要在正常子宫内膜和子宫腺肌症在位内膜腺上皮细胞核中表达,子宫腺肌症在位内膜蛋白表达高于正常子宫内膜(P<0.05),正常子宫内膜和子宫腺肌症在位内膜相应的增殖期和分泌期p-STAT3(Y705)的表达无显著性差异(P>0.05)。正常子宫内膜和子宫腺肌症在位内膜中,STAT3在mRNA和蛋白表达水平差异均无显著性(P>0.05)。VEGF主要表达在腺上皮细胞中,子宫腺肌症在位内膜蛋白表达高于正常子宫内膜(P<0.05),正常子宫内膜和子宫腺肌症在位内膜相应的增殖期和分泌期VEGF的表达差异无显著性(P>0.05)。结论 p-STAT3(Y705)促进血管生成对子宫腺肌症的发生发展有一定的作用。
Objective To investigate the localization and expression of phosphorylated signal transducers and activators of transcription 3 (Y705) and vascular endothelial growth factor (VEGF) in the eutopic endometrium of normal endometrium and adenomyosis The role of p-STAT3 (Y705) and VEGF in development of adenomyosis occurs. Methods Immunohistochemical SP method was used to detect the localization and expression of p-STAT3 (Y705) and VEGF in 14 cases of normal endometrium and 14 cases of adenomyosis eutopic endometrium. Real-time PCR was used to detect the expression of endometrial and uterine STAT3 mRNA expression in eutopic endometrium of adenomyosis; Western blotting was used to detect the expression of p-STAT3 (Y705), STAT3 and VEGF protein in eutopic and eutopic endometrium of normal endometrium and adenomyosis. Results The expression of p-STAT3 (Y705) in eutopic endometrium and normal endometrium was higher in normal endometrium and adenomyosis than in normal endometrium (P <0.05) There was no significant difference in the expression of p-STAT3 (Y705) between normal endometrium and normal endometrium in normal endometrium and normal endometrium (P> 0.05). Normal endometrium and adenomyosis eutopic endometrium, STAT3 mRNA and protein expression levels were not significantly different (P> 0.05). VEGF was mainly expressed in glandular epithelial cells, the expression of eutopic endometrium protein in adenomyosis was higher than that in normal endometrium (P <0.05), and the corresponding proliferative phase and secretion of endometrium in normal endometrium and adenomyosis There was no significant difference in the expression of VEGF (P> 0.05). Conclusion p-STAT3 (Y705) promotes angiogenesis to play a role in the development of adenomyosis.
