目的:研究人乳头瘤病毒16(HPV16)相关宫颈癌细胞系和组织中E7和肾细胞癌下调基因1(DRR1)表达的关系。方法:以HPV16 E7阳性的宫颈癌Ca Ski和Si Ha细胞、HPV16 E7阴性的宫颈癌C-33A细胞及184例宫颈鳞状细胞癌组织为研究对象,采用Western blot方法和免疫组化染色方法检测E7和DRR1蛋白的表达,分析E7与DRR1蛋白表达的关系。结果:Western blot结果显示,C-33A细胞中HPV16 E7无表达,DRR1蛋白的表达显著高于Ca Ski细胞和Si Ha细胞(F=454.982,P<0.001);Ca Ski细胞和Si Ha细胞表达HPV16 E7,二者DRR1蛋白的表达差异无统计学意义(P>0.05)。免疫组化染色结果显示,HPV16 E7阳性宫颈鳞状细胞癌组织141例,其中DRR1蛋白阳性34例;HPV16 E7阴性宫颈鳞状细胞癌组织43例,其中DRR1蛋白阳性32例;HPV16 E7阳性组织中DRR1蛋白的表达显著低于HPV16 E7阴性组织(χ2=36.250,P<0.001)。结论:DRR1可能是HPV16 E7作用的潜在靶基因。
Objective: To investigate the relationship between E7 and downregulation of renal cell carcinoma 1 (DRR1) in human papillomavirus 16 (HPV16) related cervical cancer cell lines and tissues. Methods: HPV16 E7 positive cervical cancer Ca Ski and Si Ha cells, HPV16 E7 negative cervical cancer C-33A cells and 184 cases of cervical squamous cell carcinoma tissues were studied by Western blot and immunohistochemical staining E7 and DRR1 protein expression analysis E7 and DRR1 protein expression. Results: The results of Western blot showed that HPV16 E7 was not expressed in C-33A cells and the expression of DRR1 protein was significantly higher than that of Ca Ski cells and Si Ha cells (F = 454.982, P <0.001); Ca Ski cells and Si Ha cells expressed HPV16 E7, the two DRR1 protein expression difference was not statistically significant (P> 0.05). The results of immunohistochemistry showed that there were 141 cases of cervical squamous cell carcinoma of HPV16 E7 positive, including 34 cases of DRR1 protein positive, 43 cases of HPV16 E7 negative cervical squamous cell carcinoma, of which DRR1 protein was positive in 32 cases and HPV16 E7 positive tissues DRR1 protein expression was significantly lower than HPV16 E7 negative tissue (χ2 = 36.250, P <0.001). Conclusion: DRR1 may be a potential target gene of HPV16 E7.
