目的研究脱氧胆酸对人胃癌BGC-823细胞的影响,并探讨其作用机制。方法采用细胞培养、MTT法、HE染色、Giemsa染色、Hoechst33258荧光染色、AO/EB荧光染色和流式细胞术法,检测脱氧胆酸对人胃癌BGC-823细胞生长抑制效应和凋亡效应及其机制。结果脱氧胆酸可明显抑制人胃癌BGC-823细胞的增殖,0.7 mmol/L脱氧胆酸处理48 h后抑制率达81.5%;经脱氧胆酸处理后,细胞发生皱缩、细胞体积变小、核染色质浓缩、核碎裂、细胞出芽以及凋亡小体形成等显著的凋亡特征。细胞周期检测出细胞被阻滞在G2/M期;线粒体膜电位明显下降;Annexin V-FITC/PI双染法检测出细胞凋亡率达40.35%。结论脱氧胆酸能够有效诱导人胃癌BGC-823细胞发生凋亡,且可能是通过线粒体途径诱导细胞凋亡,从而可为胃癌等相关恶性肿瘤疾病的治疗和凋亡机制的进一步研究提供重要理论基础和科学依据。
Objective To study the effect of deoxycholic acid on human gastric cancer cell line BGC-823 and its mechanism. Methods Cell growth, MTT assay, HE staining, Giemsa staining, Hoechst33258 fluorescence staining, AO / EB fluorescence staining and flow cytometry were used to detect the growth inhibitory effect and apoptosis effect of deoxycholic acid on human gastric cancer cell line BGC-823 mechanism. Results Deoxycholic acid could inhibit the proliferation of human gastric cancer cell line BGC-823. The inhibition rate reached 81.5% after treated with 0.7 mmol / L deoxycholic acid for 48 h. After deoxycholic acid treatment, the cell shrinks, the cell volume becomes smaller, Nuclear chromatin condensation, nuclear fragmentation, cell budding and apoptotic body formation and other significant apoptotic features. Cell cycle was detected in G2 / M phase; mitochondrial membrane potential decreased significantly; Annexin V-FITC / PI double staining detected apoptosis rate of 40.35%. Conclusion Deoxycholic acid can induce cell apoptosis in human gastric cancer cell line BGC-823 and induce cell apoptosis through mitochondrial pathway, which may provide important theoretical basis for further research on the treatment and apoptosis mechanism of gastric cancer and other malignant tumor.
