目的研究肾透明细胞癌(RCCC)中转化生长因子β1(TGF-β1)、细胞周期蛋白E(CyclinE)的表达与其生物学行为的关系。方法应用免疫组织化学(EliVision)法检测57例RCCC和20例癌周正常肾组织中TGF-β1和CyclinE蛋白的表达。结果①TGF-β1在RCCC中的阳性表达率为74%,高于癌周正常肾组织(35%),差异有统计学意义(P<0.05);CyclinE在RCCC中的阳性表达率为54%,高于癌周正常肾组织(25%),差异有统计学意义(P<0.05);②TGF-β1、CyclinE蛋白阳性表达率在肿瘤大小、转移有无组中,差异均有统计学意义(P<0.01);在年龄、性别组中均无统计学意义(P>0.05);③TGF-β1、CyclinE蛋白阳性表达率与RCCC病理分级、临床分期均呈正相关(P<0.01),并且随着病理分级、临床分期的升高而升高;④TGF-β1阳性表达率与CyclinE阳性表达率呈正相关(P<0.01)。结论联合检测TGF-β1、CyclinE蛋白的表达有助于反映RCCC生物学特性,可作为判断RCCC预后分子指标。
Objective To investigate the relationship between the expression of transforming growth factor β1 (TGF-β1), CyclinE and its biological behavior in renal clear cell carcinoma (RCCC). Methods The expressions of TGF-β1 and CyclinE proteins in 57 cases of RCCC and 20 cases of normal renal tissues were detected by immunohistochemistry (EliVision). Results ① The positive rate of TGF-β1 expression in RCCC was 74%, which was higher than that in normal renal tissue (35%), the difference was statistically significant (P <0.05). The positive rate of CyclinE in RCCC was 54% (P <0.05). (2) The positive rates of TGF-β1 and CyclinE protein in tumor size and metastasis group were significantly different (P <0.01). There was no significant difference in age and gender (P> 0.05). ③The positive rates of TGF-β1 and CyclinE were positively correlated with the pathological grade and clinical stage of RCCC (P <0.01) Grade, clinical stage increased and increased; ④TGF-β1 positive expression rate and CyclinE positive expression rate was positively correlated (P <0.01). Conclusions The combined detection of TGF-β1 and CyclinE protein expression is helpful to reflect the biological characteristics of RCCC and can be used as a molecular index to judge the prognosis of RCCC.
