本文构建了编码肾综合征出血热病毒核蛋白基因的真核表达载体pEFBOS/HTVNP,经限制性酶切及部分序列分析验证其插入正确。采用DEAEdextran法瞬时转染NIH3T324h后,经间接免疫荧光染色证实,NP基因可在细胞胞浆内高水平表达,此结果为进一步研究HFRSVNP诱导特异性CTL的产生打下了基础
In this paper, an eukaryotic expression vector pEFBOS / HTVNP coding for the hemorrhagic fever virus gene of hemorrhagic fever with renal syndrome was constructed and verified by restriction analysis and partial sequence analysis. After transient transfection of NIH3T324h by DEAEdextran method, it was confirmed by indirect immunofluorescence staining that the NP gene was highly expressed in the cytoplasm of the cells, which laid the foundation for further study on the induction of specific CTL by HFRSVNP
