目的探讨非小细胞肺癌患者外周血中人类肺特异性X蛋白(Lunx)mRNA的表达情况。方法选取2012年6月至2014年10月间收治的60例非小细胞肺癌患者作为研究组,选取同期健康体检人员(健康对照组)40名及肺部良性疾病者(肺部良性疾病组)30例作为对照。采用实时荧光定量反转录聚合酶链反应(FQ-RT-PCR)法定量检测患者外周血中Lunx mRNA的表达情况,并分析Lunx mRNA表达情况及其与临床病理特征的关系。结果研究组患者Lunx mRNA表达阳性者34例(56.7%)。健康对照组及肺部良性疾病组患者Lunx mRNA表达阴性。Lunx mRNA表达与研究组患者临床分期相关(P<0.05),但与患者性别、病理分型、性别及分化程度等均无关(均P>0.05)。结论应用FQ-RT-PCR法定量检测非小细胞肺癌患者外周血中Lunx mRNA敏感性及特异性均较高,从而为临床早期诊断肺癌微转移提供一定帮助,并指导临床病理分期及治疗方案的选择,改善其预后。
Objective To investigate the expression of human lung-specific X protein (Lunx) mRNA in peripheral blood of patients with non-small cell lung cancer. Methods Sixty non-small cell lung cancer patients from June 2012 to October 2014 were selected as the research group. Forty healthy subjects (healthy control group) and 40 patients with benign lung disease (benign lung disease group) 30 cases as a control. The expression of Lunx mRNA in peripheral blood of patients was quantitatively determined by real-time fluorescence quantitative reverse transcription-polymerase chain reaction (FQ-RT-PCR), and the relationship between Lunx mRNA expression and clinicopathological features was analyzed. Results In the study group, 34 cases (56.7%) had positive Lunx mRNA expression. Lunx mRNA expression was negative in healthy control group and benign lung disease group. The expression of Lunx mRNA was correlated with the clinical stage of study group (P <0.05), but not with the gender, pathological type, sex and degree of differentiation (all P> 0.05). Conclusion The quantitative and qualitative detection of Lunx mRNA in peripheral blood of patients with non-small cell lung cancer by FQ-RT-PCR has a higher sensitivity and specificity, which will be helpful for the early diagnosis of micrometastasis of lung cancer and to guide the clinicopathological stage and treatment Choose to improve their prognosis.
