目的:观察替米沙坦对胰岛素抵抗大鼠的治疗作用。方法:SD大鼠,雌雄各半,随机分为5组,每组10只。模型对照组、低剂量组、高剂量组和卡托普利组动物均给予高脂肪饲料,正常对照组给普通饲料。4周后,检测各组血脂、空腹血糖(FBG)、空腹血清胰岛素(FINS),计算胰岛素敏感性指数(ISI),评估胰岛素抵抗。正常对照组仍继续给普通饲料,其他各组继续高脂肪饲料同时,低剂量给药组灌胃替米沙坦10 mg·kg~(-1)·d~(-1)×28 d,高剂量给药组灌胃替米沙坦50 mg·kg~(-1)·d~(-1)×28 d,卡托普利组灌胃卡托普利10 mg·kg~(-1)·d~(-1)×28 d;正常对照组和模型对照组灌胃等体积0.9%氯化钠溶液。8周后检测血脂、FBG和FINS的变化,计算ISI。结果:造模阶段:造模各组FINS升高(P<0.01),ISI下降(P<0.01),但FBG无明显变化;用药阶段:替米沙坦可降低FINS,升高ISI,降低甘油三酯,均有显著差别。结论:替米沙坦对胰岛素抵抗有一定的治疗作用。
Objective: To observe the therapeutic effect of telmisartan on insulin resistance in rats. Methods: SD rats, male and female, were randomly divided into 5 groups of 10 rats. The model control group, low-dose group, high-dose group and captopril group were given high-fat diet, normal control group to ordinary feed. Four weeks later, the levels of lipids, fasting blood glucose (FBG) and fasting serum insulin (FINS) of each group were measured and the insulin sensitivity index (ISI) was calculated to evaluate the insulin resistance. The normal control group still continued to normal feed, the other groups continue to high fat feed at the same time, low dose group were given telmisartan 10 mg · kg -1 d -1 × 28 d, The rats in the captopril group were treated with captopril 10 mg · kg ~ (-1) and were given telmisartan 50 mg · kg ~ (-1) · d ~ (-1) × 28 d, · D ~ (-1) × 28 d. The normal control group and the model control group were given 0.9% sodium chloride solution. After 8 weeks, the changes of blood lipids, FBG and FINS were measured to calculate ISI. Results: In the model-making stage, FINS increased (P <0.01), ISI decreased (P <0.01), but there was no significant change in FBG. Drug treatment: telmisartan decreased FINS, increased ISI, Triester, there are significant differences. Conclusion: Telmisartan has a certain therapeutic effect on insulin resistance.
