目的在制备阳离子化小牛血清白蛋白(C-BSA)渗透泵致肾小球肾炎(CGN)大鼠模型的基础上,探讨真武汤对该模型大鼠的治疗作用。方法取雄性SD大鼠60只,其中50只大鼠埋入C-BSA渗透泵制备CGN模型,10只作为空白对照组(假手术处理)。确定造模成功后,随机分为CGN模型组,真武汤高、中、低剂量组(16.8,8.4,4.2g/kg),醋酸泼尼松组(8 mg/kg)。采用考马斯亮兰法测定24 h尿蛋白;全自动生化分析仪检测血液生化指标,包括血清总蛋白(TP)、白蛋白(ALB)、肌酐(SCr)、尿素氮(BUN)、总胆固醇(TC)和甘油三酯(TG);免疫荧光检查肾小球IgG的表达。结果与空白对照组比较,模型组大鼠24 h尿蛋白、SCr、BUN、TC和TG均明显升高(P<0.01),TP和ALB降低(P<0.01,P<0.05),且免疫荧光检查显示肾小球系膜区IgG大量沉积。与模型组比较,真武汤各剂量组大鼠24 h尿蛋白、SCr、BUN、TC和TG明显降低(P<0.01,P<0.05),TP和ALB升高(P<0.01,P<0.05),肾小球系膜区IgG沉积明显减少,尤以真武汤高剂量组效果最为显著。结论真武汤可减轻C-BSA渗透泵致CGN大鼠的肾脏免疫病理损伤,减少尿蛋白含量、改善肾功能、降低血脂,对肾脏有保护作用。
Objective To investigate the therapeutic effect of Zhenwu Decoction on rat model of glomerulonephritis (CGN) induced by cationic bovine serum albumin (C-BSA) osmotic pump. Methods Sixty male Sprague-Dawley rats were used. Fifty rats were implanted with C-BSA osmotic pump to prepare CGN model. Ten rats were used as blank control group (sham operation). After establishing the successful model, they were randomly divided into CGN model group, Zhenwu Tang high, medium and low dose groups (16.8, 8.4, 4.2g / kg) and prednisone acetate group (8 mg / kg). The 24 h urinary protein was measured by Coomassie brilliant blue method. Conclusions Zhenwu decoction can reduce renal immune pathological damage induced by C-BSA osmotic pump CGN rats, reduce urinary protein content, improve renal function, lower blood lipid and protect kidney.
