目的研究凝血酶及其受体在促进多发性骨髓瘤细胞增殖和抗凋亡作用中的机制。方法采用免疫荧光标记、流式细胞仪检测、细胞计数和免疫印迹等方法,对多发性骨髓瘤细胞株XG-1细胞表达的凝血酶受体、凝血酶对XG-1的增殖与抗凋亡作用及细胞内JAK2-STAT3分子磷酸化进行分析。结果凝血酶可以通过激活XG-1表面的凝血酶受体,继而激活JAK2-STAT3途径的磷酸化,促进骨髓瘤细胞的增殖与抗凋亡作用,且该作用与gp130介导的信号转导作用无关。结论凝血酶及其受体介导的信号转导通路是多发性骨髓瘤维持增殖和抗凋亡作用的因素之一。
Objective To study the mechanism of thrombin and its receptor in promoting the proliferation and anti-apoptosis of multiple myeloma cells. Methods The proliferation and anti-apoptotic effects of thrombin receptor and thrombin on XG-1 cells in multiple myeloma cell line XG-1 were detected by immunofluorescence staining, flow cytometry, cell counting and Western blotting.
