The authors used polymerase chain reaction (PCR) to detect the association between a novel human herpes-like virus and various types of Kaposi’s sarcoma. They collected 30 cases of Kaposi’s sarcoma specimens, 10 cases of local Kaposi’s sarcoma in Africa, 12 cases of AIDS-related patients from the United States, and 8 cases of typical Kaposi’s sarcoma. In addition, 10 cases of live fleas from normal skin of HIV-negative patients in the formative surgery were collected as controls. DNA was extracted from fresh tissues or from formalin fixed paraffin-embedded 5 μm sections for PCR amplification. The specific primer pairs are herpes virus-like DNA (5’-AGC, CGA, AAG, GAT, TCC, ACC, ATT-3’ and 5’-TCC, GTG, TTG, TCT, ACG, TCC, AGA-3’).
