目的:观察加味丹参饮(JWDSY)含药血清诱导的骨髓间充质干细胞(BMSCs),移植于心肌缺血再灌注损伤(IRI)大鼠梗死心肌边缘后,对心肌细胞碱性成纤维细胞生长因子(bFGF)蛋白表达的影响。方法:全骨髓贴壁法培养BMSCs,流式细胞术鉴定细胞表面抗原CD90,CD34,CD45。传至P3代时,用JWDSY含药血清诱导3 d。建立大鼠心肌IRI造模,在心肌梗死区边缘移植血清诱导的细胞悬液。3,7,14 d后,取心肌组织用免疫组化法检测心肌细胞bFGF表达。结果:JWDSY血清组bFGF的积分吸光度(IA)在3,7,14 d后分别升高至5 758.18±465.17,6 589.61±432.89,8 015.62±366.34;与模型组差异有统计学意义(P<0.05),且随时间逐渐升高,比较差异有统计学意义(P<0.05)。结论:JWDSY血清诱导的BMSCs移植于IRI大鼠心梗区边缘后,可以改善心肌组织的病理学形态,增加心肌细胞bFGF蛋白表达,提示JWDSY含药血清诱导的BMSCs对IRI大鼠的心肌细胞有一定的保护作用。
OBJECTIVE: To observe the effects of JWDSY-containing serum on bone marrow mesenchymal stem cells (BMSCs) transplanted into the myocardial infarcted myocardium of myocardial ischemia-reperfusion injury (IRI) Factor (bFGF) protein expression. Methods: BMSCs were cultured by whole bone marrow adherent method. The cell surface antigens CD90, CD34 and CD45 were identified by flow cytometry. To P3 generation, with JWDSY drug-containing serum induction 3 d. The establishment of rat myocardial IRI modeling, the edge of myocardial infarction area transplantation of serum-induced cell suspension. After 3, 7 and 14 days, the myocardial tissues were harvested for the detection of bFGF expression by immunohistochemistry. Results: The integral absorbance (IA) of bFGF in JWDSY serum group increased to 5 758.18 ± 465.17, 6 589.61 ± 432.89, 8 015.62 ± 366.34 respectively after 3, 7 and 14 days, and there was significant difference with model group (P < 0.05), and gradually increased with time, the difference was statistically significant (P <0.05). CONCLUSION: JWDSY serum-induced BMSCs transplantation in IRI rat myocardial infarction border can improve the pathological morphology of myocardial tissue and increase the expression of bFGF protein in myocardial cells, suggesting that JWDSY serum-induced BMSCs IRI rat myocardial cells are A certain degree of protection.
