目的:研究ω-3多不饱和脂肪酸(ω-3 PUFA)对严重烧伤大鼠肾组织及其炎性反应的影响。方法:将72只大鼠随机分成三组,即假烫组(A组)8只、对照组(中长链脂肪酸组,B组)32只和治疗组(ω-3 PUFA组,C组)32只。采用大鼠30%总体表面积(TBSA)Ⅲ度烫伤模型。B组每天按时经尾静脉注射与C组相同热量的中长链脂肪酸,C组经尾静脉注射ω-3 PUFA 2 ml/(kg·d),A组不做任何处置。分别观察A、B、C组在伤后第1、4、7和10天的血清前列腺素E2(PGE2)、白细胞介素-1(IL-1)和肿瘤坏死因子-α(TNF-α)水平。采用普通聚和酶链反应(RT-PCR)检测肾组织TNF-αmRNA的表达。各个时间点均处死8只大鼠。结果:B组和C组大鼠在伤后各时间点血清PGE2、IL-1和TNF-α水平显著高于A组(P<0.01)。C组血清PGE2、IL-1和TNF-α水平低于B组,在伤后第4、7和10天与B组比,差异有显著性统计学意义(P<0.05,P<0.01)。另外,B组和C组大鼠在伤后各时间点肾组织TNF-αmRNA表达较A组明显升高。与B组比较,C组肾组织TNF-αmRNA的表达明显降低。结论:ω-3 PUFA能降低烧伤后大鼠血清PGE2、IL-1和TNF-α水平,减轻烧伤后肾组织TNF-αmRNA的表达,可能对烧伤后肾组织具有保护作用。
Objective: To investigate the effects of ω-3 polyunsaturated fatty acids (ω-3 PUFA) on renal tissues and their inflammatory responses in severely burned rats. Methods: Seventy two rats were randomly divided into three groups, namely, fake hot group (A group), 8 control group (medium long chain fatty acid group, B group) and 32 treatment group (ω-3 PUFA group, C group) 32 only. Rat 30% total surface area (TBSA) degree scald model was used. Group B received daily oral administration of medium and long chain fatty acids with the same calorie content as group C via the tail vein on a daily basis. Group C received omega 3 PUFA 2 ml / (kg • d) via the tail vein, and group A received no treatment. The levels of serum prostaglandin E2 (PGE2), interleukin-1 (IL-1) and tumor necrosis factor-α (TNF-α) in groups A, B and C on the 1st, 4th, Level. The expression of TNF-αmRNA in renal tissue was detected by ordinary polymerase chain reaction (RT-PCR). All rats were sacrificed at each time point. Results: The levels of serum PGE2, IL-1 and TNF-α in group B and group C at each time point after injury were significantly higher than those in group A (P <0.01). The serum levels of PGE2, IL-1 and TNF-α in group C were lower than those in group B, and there was significant difference between group B and the 4th, 7th and 10th day after injury (P <0.05, P <0.01). In addition, the expression of TNF-αmRNA in kidney of group B and group C were significantly higher than that of group A at each time point after injury. Compared with group B, the expression of TNF-αmRNA in group C was significantly decreased. CONCLUSION: Omega-3 PUFA can decrease the levels of PGE2, IL-1 and TNF-α in serum and the expression of TNF-α mRNA in post-burn renal tissue, which may have a protective effect on post-burn renal tissue.
