目的观察三氧化二砷(As2O3)对体外培养的3T3成纤维细胞的增殖及分泌功能的影响。方法体外培养3T3成纤维细胞,用As2O3浓度分别为0(DMEM对照组)、2μmol/L和4μmol/L的DMEM培养液分别作用于成纤维细胞,24h和48h后MTT法观察细胞生长抑制情况,用细胞抑制率表示;As2O3各组作用细胞48h后Western blot检测I型胶原(ColI)的表达;As2O3各组作用细胞48h后放射免疫法检测上清液白细胞介素(IL)-6、IL-8的水平。结果 As2O3可显著抑制3T3成纤维细胞增殖,且呈剂量-效应关系(P<0.01)。As2O3组Ⅰ型胶原的量显著低于DMEM对照组(P<0.01),但4μmol/LAs2O3组与2μmol/LAs2O3组相比无显著性差异(P>0.05)。As2O3组IL-6和IL-8水平均显著低于DMEM对照组(P<0.01),但2μmol/L和4μmol/LAs2O3组相比差异无显著性(P>0.05)。结论 As2O3抑制3T3成纤维细胞增殖可能与抑制CoII、IL-6及IL-8的表达有关。
Objective To observe the effects of arsenic trioxide (As2O3) on the proliferation and secretion of 3T3 fibroblasts cultured in vitro. Methods 3T3 fibroblasts were cultured in vitro. The fibroblasts were treated with 0, DMEM control group, 2μmol / L and 4μmol / L As2O3 respectively, and the growth inhibition was observed by MTT assay 24h, 48h later. The cells were treated with As2O3 for 48h and the expression of type I collagen (ColI) was detected by Western blot. After treated with As2O3 for 48h, the levels of IL-6 and IL- 8 level. Results As2O3 significantly inhibited the proliferation of 3T3 fibroblasts in a dose-response manner (P <0.01). The amount of type I collagen in As2O3 group was significantly lower than that in DMEM control group (P <0.01), but there was no significant difference between 4μmol / L As2O3 group and 2μmol / L As2O3 group (P> 0.05). The levels of IL-6 and IL-8 in As2O3 group were significantly lower than those in DMEM control group (P <0.01), but there was no significant difference between 2μmol / L and 4μmol / L As2O3 groups (P> 0.05). Conclusion As2O3 inhibits the proliferation of 3T3 fibroblasts possibly by inhibiting the expression of CoII, IL-6 and IL-8.
