目的观察羧甲基壳聚糖钙对染铅小鼠体内铅、钙、锌含量及其抗氧化能力的影响。方法将一级昆明种小鼠分为正常对照组、碳酸钙对照组、染铅模型组、羧甲基壳聚糖钙低、中、高剂量实验组及其依地酸钙钠对照组,饮用醋酸铅水溶液建立小鼠预防性铅中毒模型,同时灌胃给予受试物。连续30 d后,采血,取肝脏、脑、股骨,用原子吸收分光光度计测定其铅、锌、钙含量。同时,用试剂盒测定小鼠肝组织中的丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、总抗氧化能力(T-AOC)值。结果羧甲基壳聚糖钙能使染铅小鼠体内血液、全脑、肝脏、股骨中铅含量由约1.56μg/g、13.38μg/g、16.15μg/g、1011.62μg/g显著降低至0.50μg/g、5.57μg/g、5.64μg/g、457.86μg/g,并使股骨钙含量显著升高,对小鼠全血和其他组织内锌含量无影响;羧甲基壳聚糖钙能使染铅小鼠肝脏中MDA值显著降低,SOD、GSH-Px、T-AOC活性显著升高。结论羧甲基壳聚糖钙能显著促进染铅小鼠排铅,提高骨钙含量,增强抗氧化能力。
Objective To investigate the effect of carboxymethyl chitosan calcium on lead, calcium and zinc contents and antioxidant capacity in lead-exposed mice. Methods The primary Kunming mice were divided into normal control group, calcium carbonate control group, lead-exposed model group, low, middle and high dose of carboxymethyl chitosan and the control group of calcium and sodium edetate, Lead acetate aqueous solution to establish a model of preventive lead poisoning in mice, while gavage given test substance. After 30 days, blood, liver, brain and femur were taken for determination of lead, zinc and calcium contents by atomic absorption spectrophotometer. At the same time, the contents of malondialdehyde (MDA), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), total antioxidant capacity (T-AOC )value. Results The results showed that the content of lead in blood, whole brain, liver and femur of lead-exposed mice was significantly decreased from about 1.56μg / g, 13.38μg / g, 16.15μg / g and 1011.62μg / g to 0.50μg / g, 5.57μg / g, 5.64μg / g, 457.86μg / g, and the femoral calcium content was significantly increased, and had no effect on the zinc content in whole blood and other tissues of mice; carboxymethyl chitosan calcium Can make the MDA of the lead-exposed mice significantly reduce the liver, SOD, GSH-Px, T-AOC activity was significantly increased. Conclusion Carboxymethyl chitosan calcium can significantly promote the lead excretion of lead-exposed mice, increase the content of calcium and enhance the anti-oxidation ability.
