目的研究表皮细胞培养膜片的冷冻保存。方法实验比较两种不同的冷冻保存方法和三种不同的冷冻保护液的效果。冷冻保存方法是：将表皮细胞培养膜片固定在含硅油纱布上进行冷冻保存（方法A）和将在培养瓶中的表皮细胞培养膜片直接冷冻保存（方法B）。冷冻保护液为：A．70mlRPMI＋10mlDMSO＋4％人体白蛋白20ml；B．40mlRPMI＋43ml甘油＋4％人体白蛋白17ml；C．87．5mlB21＋12．5mlDMSO＋3gPEG。结果采用方法B和B种冷冻保护液对表皮细胞培养膜片进行冷冻保存效果较好。结论对表皮细胞培养膜片进行冷冻保存是可行的。
Objective To study the cryopreservation of cultured epidermal cells. Methods Experiments were performed to compare the effects of two different cryopreservation methods and three different cryoprotectants. Cryopreservation methods are: Cryopreservation of epidermal cell culture membrane on silicone oil gauze (Method A) and cryopreservation of epidermal cell culture membrane in culture flasks (Method B). Cryoprotectant solution: A. 70mlRPMI + 10mlDMSO + 4% human albumin 20ml; 40mlRPMI + 43ml glycerol + 4% human albumin 17ml; 87.5 ml B21 + 12.5 ml DMSO + 3 g PEG. Results The method of cryopreservation of B and B kinds of cryoprotective fluid on epidermal cell culture membrane cryopreservation better. Conclusion It is feasible to cryopreserve the epidermal cell culture membrane.
