为探讨复方苦豆子(CK)对肾性高血压大鼠(RHR)左室肥厚(LVH)及血浆血管紧张素II(AngII)的影响,建立了二肾一夹型高血压大鼠模型(2KⅠC-RHR),选取造模成功的大鼠57只,随机分为5组:模型组(Mod组),CK高、中、低剂量组(H、M、L组)和卡托普利组(Cap组),另取10只作为假手术组(Sham组)。每日灌药1次,连续8周。根据体重随时调整给药量。观察药物对模型动物左室肥厚指数(LVHI)及AngⅡ影响。结果表明:1)CK长期给药能明显降低模型动物的LVHI,H、M组与Mod组相比均有显著性差异(P<0.01),L组也有下降趋势,但无统计学意义。2)CK能降低模型动物血浆AngⅡ水平,与Mod组相比,各给药组均有显著性差异(P<0.01),以H组下降最为明显。这表明CK对RHR的心脏重塑有一定的防治作用,其机制可能与抑制肾素-血管紧张素系统(RAS)活性有关。
To investigate the effect of compound Kudouzi (CK) on left ventricular hypertrophy (LVH) and plasma angiotensin II (AngII) in renal hypertensive rats (RHR), a two-kidney, one-clip hypertensive rat model was established (2KIC -RHR). 57 rats were randomly selected and randomly divided into 5 groups: model group (Mod group), CK high, middle and low dose group (H, M, L group) and captopril group ( Cap group), another 10 were taken as a sham group (Sham group). Dosing once daily for 8 weeks. The dosage was adjusted at any time according to body weight. Observed the effect of drugs on left ventricular hypertrophy index (LVHI) and AngII in model animals. The results showed that: 1) Long-term administration of CK can significantly reduce LVHI in model animals, and there is a significant difference between H and M groups compared with Mod group (P<0.01). L group also has a decreasing trend, but it has no statistical significance. 2) CK can reduce plasma Ang II levels in model animals. Compared with Mod group, each group has significant difference (P<0.01). The decrease is most obvious in group H. This indicates that CK has a certain preventive effect on cardiac remodeling of RHR, and its mechanism may be related to the inhibition of renin-angiotensin system (RAS) activity.
