目的探讨对羟基苯甲酸丙酯对人角质形成细胞(HaCat)的细胞毒作用。方法选用人角质形成细胞株,分别观察不同剂量(3.125~200μg/ml)对羟基苯甲酸丙酯作用不同时间(24~72h)对其细胞毒作用。通过MTT比色法测定其对细胞增殖活力的影响;乳酸脱氢酶(LDH)释放法测定其对细胞膜的损伤。结果对羟基苯甲酸丙酯可引起培养细胞数量明显减少,随作用时间的延长,剂量的增加,对角质形成细胞增殖的抑制作用也显著增大,作用24h在50μg/ml出现细胞增殖抑制,作用48h在25μg/ml出现细胞增殖抑制,而作用72h在6.25μg/ml时就出现细胞增殖抑制,呈现明显的时间-剂量-效应关系;与对照组比较,从50μg/ml起,LDH释放率明显增加,并随剂量的增加而呈上升趋势。结论对羟基苯甲酸丙酯对HaCat细胞具有明显的细胞毒作用,呈现明显的时间-剂量-效应关系。
Objective To investigate the cytotoxicity of propylparaben on human keratinocytes (HaCat). Methods Human keratinocyte cell line was used to observe the cytotoxicity of propylparaben (3.125 ~ 200μg / ml) at different times (24 ~ 72h). MTT colorimetric method was used to determine the effect on cell proliferation and lactate dehydrogenase (LDH) release assay. Results Propylparaben could significantly reduce the number of cultured cells. With the extension of time and dosage, the inhibitory effect on the proliferation of keratinocytes was also significantly increased. The cell proliferation was inhibited at 50μg / ml for 24h 48h at 25μg / ml cell proliferation inhibition, and the role of 72h at 6.25μg / ml appeared cell proliferation inhibition, showing a significant time-dose-effect relationship; compared with the control group, from 50μg / ml LDH release rate was significantly Increase, and with the increase of dose is on the rise. Conclusion Propylparaben showed obvious cytotoxic effect on HaCat cells and showed a significant time-dose-response relationship.
