目的 研究氧化苦参碱与国产α lb和α 2b干扰素对大鼠星状细胞增殖和胶原合成的影响 ,为应用氧化苦参碱防治肝纤维化提供理论依据。方法 分离、培养大鼠星状细胞 ,采用MTT法和3 H 脯氨酸掺入法分别测定细胞增殖和胶原合成。结果 氧化苦参碱在 5~ 5 0 0 μg·ml-1浓度范围内、2种α 干扰素在 5 0 0~ 10 0 0 0U·ml-1浓度范围内能呈剂量依赖性抑制星状细胞的增殖和胶原合成 ,提高药物浓度则形成作用平台。结论 氧化苦参碱与α 干扰素在体外均具有抑制大鼠星状细胞增殖和胶原合成的作用 ,这对防治肝纤维化可能有一定的临床应用价值。
Objective To study the effects of oxymatrine and domestic α lb and α 2b interferon on the proliferation and collagen synthesis of rat stellate cells, and provide theoretical basis for the prevention and treatment of liver fibrosis by oxymatrine. Methods Rat stellate cells were isolated and cultured, and cell proliferation and collagen synthesis were measured by MTT method and 3 H proline incorporation method. Results Oxymatrine dose-dependently inhibited stellate cells in the concentration range of 5-500 μg·ml-1 and two kinds of interferon α in the range of 500-1000 U·ml-1. The proliferation and collagen synthesis, and increasing drug concentrations form a role platform. Conclusion Oxymatrine and α-interferon can inhibit the proliferation and collagen synthesis of rat stellate cells in vitro, which may have certain clinical application value in the prevention and treatment of liver fibrosis.
