目的:观察养心通脉有效部位方(apr-YTF)对大鼠骨髓间充质干细胞(BMSCs)归巢梗死心肌的影响。方法:将急性心肌梗死大鼠模型随机分成apr-YTF注射液、rhG-CSF注射液和PBS缓冲液3组,测量各组大鼠干预后在体心功能、心室肌瘢痕面积、瘢痕毛细血管数、心肌CD34+细胞、Brdu与cTn I双染阳性细胞数、Ⅷ因子、VEGF、bFGF、Flk-1、外周血CD34+细胞数,分析组间差异。结果:①与PBS组比较,apr-YTF组和rhG-CSF组的LVSP、LVEDP、+dp/dtmax、-dp/dtmax均有显著改善,心室肌瘢痕面积显著减小,心室肌毛细血管数、心肌边缘区和外周血液CD34+染色阳性细胞数、心肌梗死边缘区中Brdu与cTn I双染阳性细胞数及Ⅷ因子、Flk-1、VEGF、bFGF的表达均显著增加(P<0.01,P<0.05);②与rhG-CSF组比较,apr-YTF组的LVSP显著升高,心肌组织CD34+染色阳性细胞数、心肌梗死边缘区中Brdu与cTn I双染阳性细胞数均显著增高(P<0.01)。结论:apr-YTF能促使BMSCs动员入血、定向归巢并转化为心肌细胞,且优于rhG-CSF。
Objective: To observe the effect of apr-YTF on the infarction myocardium of rat bone marrow mesenchymal stem cells (BMSCs) homing. Methods: Acute myocardial infarction rat models were randomly divided into apr-YTF injection, rhG-CSF injection and PBS buffer 3 groups were measured in each group after the intervention in the body and heart function, ventricular muscle scar area, scar capillaries , Myocardial CD34 + cells, Brdu and cTn I double staining positive cells, factor Ⅷ, VEGF, bFGF, Flk-1, CD34 + cells in peripheral blood. Results: ①Compared with PBS group, the LVSP, LVEDP, + dp / dtmax and -dp / dtmax of apr-YTF group and rhG-CSF group were significantly improved, the area of ventricular muscle scar significantly decreased, the number of ventricular capillary, The number of positive cells of CD34 +, the number of positive Brdu and cTn I staining and the expressions of factor Ⅷ, Flk-1, VEGF and bFGF in borderline and peripheral blood of myocardium were significantly increased (P <0.01, P <0.05 ); ② Compared with rhG-CSF group, LVSP in apr-YTF group was significantly increased, the number of CD34 + positive cells in myocardial tissue and the number of Brdu and cTn I double positive cells in border area of myocardial infarction were significantly increased (P <0.01) . Conclusion: apr-YTF can promote the mobilization of BMSCs into the blood, homing and conversion into cardiomyocytes, and is superior to rhG-CSF.
