用制备的电极测试提纯的 TMV病毒 ,在 p H6 .0 ,6 .5,7.0 ,7.5,0 .0 1mol/ L PBS缓冲液中平衡得电极 ,用对应 p H的 TMV(0 .1mg/ m L)测试 ,电极电位都增大 ,且随 p H升高 ,增大幅度减小 ;在 p H8.0 ,8.5PBS缓冲液中平衡好的电极 ,同上测试 ,电极电位下降。电极在 p H2 .8,0 .2 mol/ L甘氨酸 -盐酸缓冲液中再生后可重复使用 ,电极至少可重复使用一月。用珊西烟 TMOV病叶的粗汁液与健康叶粗汁液测试 ,前者电位变化明显比后者大 ,说明粗汁液对电位有影响 ,但不会掩盖由于免疫反应引起的电位变化
The purified TMV virus was tested using the prepared electrodes and equilibrated in p H6 .0, 6.5, 7.0, 7.5, 0.0 1 mol / L PBS buffer with TMV (0.1 mg / m L) test, the electrode potentials all increased, and the increasing amplitude decreased with the increase of p H; the well-balanced electrodes in p H8.0 and 8.5 PBS buffer were tested as above and the electrode potential decreased. The electrode can be reused after it is regenerated in p H2 .8,0 .2 mol / L glycine-HCl buffer and the electrode can be reused at least for January. With the Shanxi tobacco TMOV diseased leaf crude juice and healthy leaf crude juice test, the former potential changes were significantly larger than the latter, indicating that crude juice on the potential impact, but will not mask the potential changes due to immune response
