目的研究膀胱癌患者尿沉渣中miR-142-3p和miR-145-5p差异性表达,为临床寻找早期膀胱癌肿瘤标志物提供依据。方法搜集51例未接受经尿道肿瘤切除术的早期膀胱癌患者和38例健康对照者的清晨中后段尿液,从尿沉渣中提取RNA,采用miRNA特异性茎环逆转录荧光定量PCR检测目标miRNA表达水平,分析两组的受试者工作曲线(ROC),比较miR-142-3p和miR-145-5p及其组合在膀胱癌早期诊断中的价值。结果 miRNA-142-3p在早期膀胱癌患者尿沉渣中显著升高(FC=3.93,P=0.0002,AUC=0.736)。组合测定miR-145-5p/miR-142-3p比值在膀胱癌患者与健康对照者间有统计学差异(P=0.00004,AUC=0.755)。结论测定尿沉渣中的miR-142-3p能有效发现早期膀胱癌,而检测miR-145-5p/miR-142-3p比值的诊断价值更高。
Objective To study the differential expression of miR-142-3p and miR-145-5p in urinary sediment in patients with bladder cancer, and to provide evidence for the clinical search of early stage bladder cancer tumor markers. Methods 51 early urinary bladder cancer patients without early urethral resection and 38 healthy controls were enrolled in this study. RNA was extracted from urinary sediment and miRNA-specific stem-loop RT-PCR was used to detect the target miRNA expression levels, receiver operating characteristic curve (ROC) of the two groups were analyzed to compare the value of miR-142-3p and miR-145-5p and their combination in the early diagnosis of bladder cancer. Results miRNA-142-3p was significantly increased in urinary sediment in patients with early stage bladder cancer (FC = 3.93, P = 0.0002, AUC = 0.736). The combination of miR-145-5p / miR-142-3p ratio in patients with bladder cancer and healthy controls were statistically significant (P = 0.00004, AUC = 0.755). Conclusions The determination of miR-142-3p in urine sediment can effectively detect early stage bladder cancer, and the diagnostic value of miR-145-5p / miR-142-3p is higher.
