目的分析阿魏菇和杏鲍菇2种侧耳多糖的单糖组成。方法阿魏菇多糖和杏鲍菇多糖经完全酸水解后,用薄层色谱法(thin layer chromatography,TLC)和气相色谱法(gas chromatography,GC)分析样品的单糖组成,并测定各单糖的摩尔比例。结果 TLC法和GC法均检测出阿魏菇多糖和杏鲍菇多糖含有阿拉伯糖(Ara)、甘露糖(Man)、葡萄糖(Glc)和半乳糖(Gal)单糖组分,GC法测得阿魏菇多糖和杏鲍菇多糖中各单糖的摩尔比(Ara:Man:Glc:Gal)分别为1.00:1.75:11.79:2.22和1.00:4.53:1.92:2.24。结论阿魏菇多糖和杏鲍菇多糖的单糖组成类似,但二者的各单糖摩尔比例有差异,TLC和GC法适用于分析阿魏菇和杏鲍菇多糖的单糖组成。
Objective To analyze the monosaccharide compositions of Pleurotus ostreatus and Pleurotus eryngii. Methods The polysaccharides of Pleurotus ostreatus and Pleurotus eryngii were completely acid-hydrolyzed. The monosaccharide compositions of the samples were analyzed by thin layer chromatography (TLC) and gas chromatography (GC) The molar ratio. Results The contents of Ara, Man, Glc and Gal monosaccharides in both Pleurotus ostreatus and Pleurotus eryngii polysaccharides were detected by TLC and GC. The molar ratios (Ara: Man: Glc: Gal) of the polysaccharides of Pleurotus ferulae and Pleurotus eryngii were 1.00: 1.75: 11.79: 2.22 and 1.00: 4.53: 1.92: 2.24, respectively. Conclusion The polysaccharides of Pleurotus ostreatus and Pleurotus eryngii polysaccharide have similar monosaccharide composition, but the molar ratio of the two polysaccharides is different. The TLC and GC methods are suitable for the analysis of the monosaccharide composition of Pleurotus ferulae and Pleurotus eryngii polysaccharides.
