目的探讨熊果酸对胃癌细胞株SGC-7901和MGC-803增殖、凋亡的调控作用。方法SGC-7901和MGC-803细胞接种于含10%胎牛血清的RPMI 1640培养液中,常规培养24h后加熊果酸(10、20、40、80μmol/L),继续培养12、24、48h。MTT比色分析细胞增殖;药物作用24h后,流式细胞术检测细胞凋亡率;Western blot检测增殖细胞核抗原(PCNA)和前体半胱天冬氨酸蛋白酶3(pro-Caspase-3)的表达。结果熊果酸呈时间和剂量依赖性抑制SGC-7901和MGC-803细胞增殖;熊果酸剂量依赖性诱导SGC-7901和MGC-803细胞凋亡,抑制PCNA和pro-Caspase-3蛋白表达。结论熊果酸抑制SGC-7901和MGC-803细胞增殖并诱导其凋亡。其作用可能与抑制PCNA表达和诱导pro-Caspase-3活化有关。
Objective To investigate the regulation of ursolic acid on the proliferation and apoptosis of gastric cancer cell lines SGC-7901 and MGC-803. Methods SGC-7901 and MGC-803 cells were inoculated into RPMI 1640 medium containing 10% fetal bovine serum. UA (10, 20, 40 and 80μmol / L) 48h. MTT colorimetric analysis of cell proliferation; 24h after treatment, the apoptotic rate was detected by flow cytometry; Western blot detection of proliferating cell nuclear antigen (PCNA) and pro-caspase-3 expression. Results Ursolic acid inhibited the proliferation of SGC-7901 and MGC-803 cells in a time and dose-dependent manner. UA inhibited the apoptosis of SGC-7901 and MGC-803 cells in a dose-dependent manner and inhibited the expression of PCNA and pro-Caspase-3. Conclusion Ursolic acid inhibits the proliferation and induces the apoptosis of SGC-7901 and MGC-803 cells. Its role may be related to inhibition of PCNA expression and induction of pro-Caspase-3 activation.
