用免疫组织化学方法研究了大白鼠肝脏在46℃热休克30min后恢复期(2.5~72hr)内HSP70的表达定位和动态变化及不同温度(42、44和46℃)处理30min、恢复12hr后HSP70的表达。结果表明:(1)HSP70主要定位于肝细胞的细胞质中;(2)在肝小叶中,阳性细胞多集中于中央静脉周围;(3)46℃处理30min、恢复8~12hr,HSP70表达量最多;(4)不同温度热休克处理30min、恢复12hr表明,46℃处理组诱导大白鼠肝细胞表达HSP70最多。用体视学方法对实验组的HSP70表达进行了定量组织学分析。
Immunohistochemistry was used to study the expression localization and dynamic changes of HSP70 in the livers of rats after heat shock at 46 ℃ for 30min (2.5 ~ 72hr) and the changes of HSP70 expression at different temperatures (42, 44 and 46 ℃) for 30min and 12hr After HSP70 expression. The results showed that: (1) HSP70 mainly localized in the cytoplasm of hepatocytes; (2) in the hepatic lobules, positive cells were mostly concentrated around the central vein; (3) the expression of HSP70 was up to 8 ~ 12hr at 46 ℃ for 30 ~ ; (4) heat shock treatment at different temperatures for 30min and recovery for 12hr indicated that the HSP70 expression in the treated group at 46 ℃ was the highest. Histopathological methods were used to quantitatively analyze the expression of HSP70 in the experimental group.
