目的:检测PTEN蛋白和PCNA蛋白在胃癌组织中的表达,并研究其临床病理意义。方法:用免疫组织化学S—P法检测PTEN和PCNA蛋白在80例胃癌组织中的表达,并取30例同期胃炎黏膜标本作为对照。结果:胃炎黏膜PTEN蛋白阳性表达表达率为100.0%(30/30),胃癌组织PTEN蛋白阳性表达率为40.0%(32/80);高、中分化腺癌的PTEN蛋白阳性表达率高于低分化腺癌的;Ⅰ期、Ⅱ期胃癌的PTEN蛋白阳性表达率高于Ⅲ期、Ⅳ期的;无淋巴结转移组胃癌的PTEN阳性表达率高于淋巴结转移组的;胃癌组织PCNA标记指数为(86.7±12.3)%,高于胃炎黏膜的PCNA标记指数;PTEN蛋白表达阴性的胃癌细胞PCNA的标记指数高于PTEN蛋白表达阳性者。胃癌患者生存年限>5aPTEN蛋白阳性表达率高于生存年限<5a者。结论:PTEN蛋白的表达缺失与胃癌的发生、分化、转移和预后密切相关,胃癌PTEN蛋白表达与PCNA表达呈负相关。
Objective: To detect the expression of PTEN protein and PCNA protein in gastric cancer and to study the clinicopathological significance. Methods: The expressions of PTEN and PCNA protein in 80 cases of gastric carcinoma tissues were detected by immunohistochemical S-P method, and 30 cases of gastritis mucosa samples were taken as control. Results: The positive expression rate of PTEN protein in gastritis mucosa was 100.0% (30/30), while the positive expression rate of PTEN protein in gastric carcinoma tissues was 40.0% (32/80). The positive rates of PTEN protein in high and moderately differentiated adenocarcinomas were higher than those in low The positive expression rate of PTEN protein in stage Ⅰ and Ⅱ gastric cancer was higher than that in stage Ⅲ and Ⅳ. The positive rate of PTEN in gastric cancer without lymph node metastasis was higher than that in lymph node metastasis. The index of PCNA in gastric cancer was ( 86.7 ± 12.3)%, which was higher than that of the gastric mucosa. The labeling index of PCNA in PTEN-negative gastric cancer cells was higher than that in PTEN-positive ones. Survival of patients with gastric cancer> 5aPTEN protein expression was higher than the survival of <5a. Conclusion: The loss of PTEN protein expression is closely related to the occurrence, differentiation, metastasis and prognosis of gastric cancer. The expression of PTEN in gastric cancer is negatively correlated with the expression of PCNA.
