探讨骨髓间充质干细胞在器官移植中记忆性T淋巴细胞功能的影响。通过同种异基因皮肤移植的方法诱导CD8~+记忆性T淋巴细胞的产生,在体外应用混合淋巴增殖实验观察骨髓间充质干细胞对经过刺激后的T细胞增殖情况的影响;另一方面,通过同种异基因小鼠心脏移植模型的建立,在体内验证和观察骨髓间充质干细胞对小鼠移植器官生存寿命的影响。骨髓间充质干细胞在混合淋巴增殖实验中,可以有效抑制CD8~+记忆性T淋巴细胞的增殖能力,实验组淋巴细胞增殖指数明显低于对照组(t=4.575,p<0.05);在小鼠心脏移植模型中,输注骨髓间充质干细胞后移植心脏的生存寿命明显增加,差异有统计学意义(p<0.05)。骨髓间充质干细胞能够有效抑制器官移植中CD8~+记忆性T淋巴细胞的增殖,诱导免疫耐受,延长异体器官存活时间。
To investigate the effect of bone marrow mesenchymal stem cells on memory T lymphocyte function in organ transplantation. Through allogeneic skin transplantation method to induce the generation of CD8 ~ + memory T lymphocytes in vitro mixed lymphocyte proliferation experiment was used to observe the effects of bone marrow mesenchymal stem cells on the proliferation of T cells after stimulation; the other hand, Through the establishment of allogeneic mouse heart transplantation model, the effect of bone marrow mesenchymal stem cells on the survival life of mice transplanted organ was verified and observed in vivo. In mixed lymphoproliferation experiments, BMSCs can effectively inhibit the proliferation of CD8 ~ + memory T lymphocytes. The lymphocyte proliferation index of experimental group was significantly lower than that of control group (t = 4.575, p <0.05) In the mouse heart transplantation model, the survival life of transplanted heart after transfusion of bone marrow mesenchymal stem cells was significantly increased, the difference was statistically significant (p <0.05). Bone marrow mesenchymal stem cells can effectively inhibit the proliferation of CD8 ~ + memory T lymphocytes in organ transplantation, induce immune tolerance and prolong the survival time of allogeneic organ.
