目的:阐明5α-还原酶II(SRD5AII)在附睾上皮细胞系PC1和DC2中的表达特征。方法:应用实时荧光定量PCR法和免疫荧光法,检测附睾上皮细胞系PC1和DC2中SRD5AIImRNA的表达水平和蛋白定位。结果:附睾上皮细胞系PC1和DC2内均有SRD5AIImRNA表达,且在DC2中表达较PC1中高。SRD5AII蛋白主要定位于胞质内。结论:附睾上皮细胞系PC1和DC2内有SRD5AIImRNA和蛋白表达,提示其可能参与附睾中精子成熟的调节。
Objective: To elucidate the expression of 5α-reductase II (SRD5AII) in epididymal epithelial cell lines PC1 and DC2. Methods: Real-time fluorescence quantitative PCR and immunofluorescence were used to detect the expression of SRD5AII mRNA and protein in epididymal epithelial cell line PC1 and DC2. Results: The expressions of SRD5AII mRNA in epididymal epithelial cell lines PC1 and DC2 were both higher than those in PC1. SRD5AII protein mainly located in the cytoplasm. Conclusion: SRD5AII mRNA and protein expression in epididymal epithelial cell lines PC1 and DC2 suggest that they may participate in the regulation of sperm maturation in epididymis.
