目的探讨白细胞介素-17(IL-17)及其抗体对哮喘小鼠肺中嗜酸性粒细胞(EO S)的影响。方法随机将60只Balb/c雌性小鼠分为健康组(A)、对照组(B)、哮喘组(C)、IL-17抗体组(D)及IL-17组(E)各12只。A组不做特殊处理,B、C、D、E组采用卵蛋白致敏和激发的方法,建立哮喘模型并给予相应干预;采用HE染色肺组织病理切片观察炎性细胞浸润,采用ELISA法检测小鼠肺组织匀浆上清液中嗜酸性粒细胞趋化因子(Eotax in-1)。结果 C、D、E组小鼠气道炎症以EO S和中性粒细胞浸润为主,B组变化较轻,A组正常;Eotax in-1浓度从高到低依次为C、D、E、B、A组(P<0.05),其差异有统计学意义。结论外源性IL-17能够减少激发状态下哮喘小鼠肺中EO S的浸润。
Objective To investigate the effect of interleukin-17 (IL-17) and its antibody on eosinophil (EOS) in the lungs of asthmatic mice. Methods Sixty Balb / c female mice were randomly divided into healthy group (A), control group (B), asthma group (C), IL-17 antibody group (D) and IL-17 group . The rats in group A, B, C, D and E were sensitized and challenged with ovalbumin to establish the model of asthma and the intervention was given accordingly. Inflammatory cell infiltration was observed by HE staining and pathological examination was performed by ELISA Eotaxin-1 in mouse lung homogenate supernatant. Results The airway inflammation of mice in groups C, D and E was mainly infiltrated with EO S and neutrophil. The changes in group B were mild and normal in group A. The concentrations of Eotax in-1 were C, D, E , B, A group (P <0.05), the difference was statistically significant. Conclusion Exogenous IL-17 can reduce the infiltration of EO S in the lungs of asthmatic mice under excitation.
