本文介绍了一种简便易行的人乳头瘤病毒(HPV)快速检测与分型新方法:多聚酶链反应—酶联免疫吸附试验(PCR-ELISA)。作者采用了源自HPVL_1区的高度保守序列的通用引物对MY09[5’-CGTCC(AC)A(AG)(AG)GGA(AT)ACTGATC]/MY11[5’-GC(AC)CAGGG(AT)CATAA(CT)AATGG];并采用了2条组特异性探针GP1/GP2和5条型特异性探针:针对6型的MY12、针对11型的MY13、针对16型的MY14、针对18型的WD74和针对33型的MY16,以上探针均用生物素标记。使用德国Boehringer Mannheim公司提供的PCR-ELISA试剂盒,先以通用引物MY09/MY11和经地高辛甙元标记的dUTP进行PCR,获
This article describes a simple and easy method for the rapid detection and typing of human papillomavirus (HPV): polymerase chain reaction-enzyme-linked immunosorbent assay (PCR-ELISA). The authors used a universal primer pair MY09 [5’-CGTCC (AC) A (AG) (AG) GGA (AT) ACTGATC] / MY11 [5’-GC (AC) CAGGG ) CATAA (CT) AATGG]; and two panel-specific probes GP1 / GP2 and 5 bar-specific probes were used: MY12 for type 6, MY13 for type 11, MY14 for type 16, Type WD74 and MY16 type 33, the above probes are labeled with biotin. PCR-ELISA kit provided by Boehringer Mannheim Company of Germany was used to carry out PCR with universal primers MY09 / MY11 and digoucosidase-labeled dUTP
