目的探讨KAI1/CD82基因在人鼻咽癌细胞株及鼻咽部组织中表达水平的差异。方法应用PCR法检测5种人鼻咽癌细胞株、28例鼻咽癌和15例非肿瘤性鼻咽部组织KAI1/CD82 mRNA的表达情况。结果不同转移习性的人鼻咽癌细胞均扩增出KAI1/CD82 mRNA,随细胞转移习性增高,KAI1/CD82 mRNA表达水平降低;KAI1/CD82 mR-NA在鼻咽癌组织中的阳性表达率为42.9%,低于非肿瘤性鼻咽组织的73.3%,两者差异无统计学意义;KAI1/CD82mRNA在鼻咽癌组织中的表达与患者的性别、年龄、病理类型、T分期等无相关性。随N分期进展,KAI1/CD82 mRNA阳性表达率逐渐降低。无淋巴结转移组(N0)85.7%比有淋巴结转移组(N1-N3)28.6%表达明显增高,两者差异有统计学意义。结论 KAI1/CD82基因在低转移性人鼻咽癌细胞株中高表达,而在高转移性人鼻咽癌细胞株中低表达。KAI1/CD82基因在鼻咽癌组织中低表达,在非肿瘤性鼻咽部组织中高表达,提示该基因在鼻咽癌的发生、发展中可能起到一定作用。KAI1/CD82基因在鼻咽癌组织的低表达与鼻咽部淋巴结转移有关,提示该基因在抑制鼻咽癌转移的过程中起重要作用。
Objective To investigate the difference of KAI1 / CD82 gene expression in human nasopharyngeal carcinoma cell lines and nasopharyngeal tissues. Methods The expression of KAI1 / CD82 mRNA in 5 human nasopharyngeal carcinoma cell lines, 28 nasopharyngeal carcinoma and 15 non-neoplastic nasopharyngeal tissues were detected by PCR. Results KAI1 / CD82 mRNA was amplified in human nasopharyngeal carcinoma cells with different metastatic habits. The expression of KAI1 / CD82 mRNA was decreased with the increase of cell metastatic habits. The positive expression rate of KAI1 / CD82 mR-NA in nasopharyngeal carcinoma was 42.9%, lower than non-neoplastic nasopharyngeal tissue 73.3%, the difference was not statistically significant; KAI1 / CD82mRNA expression in nasopharyngeal carcinoma patients with gender, age, pathological type, T staging no correlation . With the progression of N stage, the positive rate of KAI1 / CD82 mRNA decreased gradually. There was a significant difference between the two groups (85.7%, 28.6%, N1-N3) in the group without lymph node metastasis (N0). Conclusions KAI1 / CD82 gene is highly expressed in low metastatic human nasopharyngeal carcinoma cell lines and low in highly metastatic human nasopharyngeal carcinoma cell lines. The low expression of KAI1 / CD82 gene in nasopharyngeal carcinoma tissues and high expression in non-neoplastic nasopharyngeal tissues suggest that this gene may play a role in the occurrence and development of nasopharyngeal carcinoma. The low expression of KAI1 / CD82 in nasopharyngeal carcinoma and nasopharyngeal lymph node metastasis, suggesting that the gene plays an important role in the inhibition of nasopharyngeal carcinoma metastasis.
