一、试验目的:探索用红四氮唑测定大麦种子生活力的判断标准。二、材料和方法:取具有不同发芽率的“沪麦4号”、“77—130”、“沪辐48”等大麦种子,用30℃温水恒温浸种4小时或于20℃左右的室温下浸种16小时,使种子软化,用解剖刀沿胚部正中将种子纵切两半,一半放入染色板小孔内,以0.2%四唑溶液在25℃左右室温下染色5~6小时,另一半对应地放入以棉花做发芽床的染色板内,在25℃左右的室温下经24小时,使其萌动,逐粒对照,观察发芽种子与染色种子的情况,判断种子是否有生活力的染色标准。三、试验结果:试验共做十一次,计种子550粒,因染色失败或发芽不好等原因,染上色又发芽
